I always interpreted Kirk’s reaction here as “only every day since I met you, oh wait, I can’t actually say that.”  He goes all shifty eyes and everything.

Also, Spock uses this weirdly expectant tone, like he’d be surprised if Jim HADN’T wondered (i.e. “I’ve seen you checking me out, man”), and Jim looks kind of put out and overly thoughtful (i.e. “I guess I never stood a chance, did I?”), and AKDJFWIHAKJHFDQIK.
